The main factors affecting the determination of jute with vegetables by chlorophyll detector

During the single factor extraction of jute chlorophyll in vegetables, the extraction efficiency of chlorophyll was significantly different among different extraction solvents, especially for chlorophyll detectors, which may be due to the interference of acetone with chlorophyll by other ingredients contained in jute. The extraction effect of the follow-up test is necessary for further analysis. The extraction efficiency is positively correlated with the extraction time. When it reaches 4 h, the chlorophyll reaches a diffusion equilibrium state and the extraction efficiency is the highest.

Pick up the leaves of the same growing Fongong No.1, clean, dry and chopped, and then take 5g each, add the corresponding organic extract 50mL, put it aside for 1min, turn into a brown bottle, choose different according to the chlorophyll detector test requirements The extraction agent, different temperatures, different extraction time for extraction. In the solid-to-liquid ratio test, the volume of the extracted solvent was taken to 50 mL, and the weight of the leaf was increased or decreased according to the ratio of the material to the liquid, and stored as a dark liquid for the chlorophyll detector to be protected from light.

At room temperature (25 °C), the lowest extraction efficiency of chlorophyll was 1.903 mg·g-1. With the increase of temperature, the extraction amount was increased by chlorophyll detector, but the increase slowed down. The main reason is that chlorophyll is not stable at 70°C, and it is easily decomposed, which causes the chlorophyll content in the extract to decrease. The smaller the ratio of material to liquid was, the smaller the ratio was, and the higher the extraction efficiency was. With the increase of extraction temperature, the extraction effect of chlorophyll was better. Considering the stability of chlorophyll and the cost factor, the optimal temperature was 40-60°C.

The chlorophyll detector was used to determine the extraction process of jute chlorophyll by three factors: feed-liquid ratio, extraction temperature and extraction time. The results showed that the main factors influencing the determination of chlorophyll detector and chlorophyll extraction were: extraction Temperature> solid-liquid ratio> extraction time, and the influence of the ratio of material to liquid, extraction temperature, and extraction time all reached a very significant level.
